Sulfo-NHS-SS-biotin是一种胺反应性的生物素化试剂。Sulfo-NHS-SS-biotin含有带负电荷的磺酸基,使其分子内产生足够的极性,可以直接加入到反应性水溶液中,而不需要提前溶解在有机溶剂中。尽管不需要提前溶解,sulfo-NHS-SS-biotin的水溶性原液必须快速制备并立即使用,以防止活性酯的水解。Sulfo-NHS-SS-biotin与含胺的蛋白或其它分子反应形成一个复合体,该复合体进一步与亲和素或链霉素探针相互作用,用亲和层析的方法纯化目标分子。

• Medium length—spacer arm is 24.3 angstroms; it consists of the native biotin valeric acid group extended by a 7-atom chain

应用 |
细胞悬液(1.5×106)用预冷的PBS洗三次后与1mg/mL sulfo-NHS-SS-biotin混合,在冰上放置15分钟。用含100 mmol/L谷氨酸的PBS能够终止生物素化反应。细胞经过PBS清洗后,用含蛋白酶抑制剂的RIPAh缓冲液(20 mmol/L sodium phosphate, 150 mmol/L NaCl (pH 7.4), 1% NP40, 0.1% SDS, and 0.5% deoxycholic acid)重悬。生物素化的膜蛋白用链霉亲和素琼脂糖拉下来,用SDS样品缓冲液洗脱出来,经过SDS-PAGE,电转到PVDF膜上, 最后用抗体进行杂交。 |
