Glimepiride是一种磺酰脲类化合物,用于控制2型糖尿病病人的血糖水平。
当培养的细胞在生理学剂量的胰岛素和glimepiride(10 μM)存在时,2-脱氧葡萄糖的摄取增加到对照组的186%。Glimepiride在胰岛素不存在的情况下也能提高2-脱氧葡萄糖的摄取。同时,glimepiride分别将GLUT1和GLUT4的表达量提高为对照组的164%和148%。这些结果表明,glimepiride可通过不依赖胰岛素的通路增加心脏葡萄糖摄取[1]。
在糖尿病易发(DP)BB大鼠中,Glimepiride(200 mg/kg/日)使糖尿病发病率相对于对照组降低了23%[2]。在高胰岛素血症高血糖KK-Ay小鼠中,glimepiride使血糖减少了40%,HBA1c减少了33%,血浆胰岛素减少了50%。在大鼠隔膜和3T3脂肪细胞中,在胰岛素缺乏的情况下,glimepiride使葡糖糖转运和最大胰岛素反应分别增加到60%和35%[3]。
