爱拉斯汀(Erastin)是细胞渗透性ferroptosis的激活剂和抗肿瘤剂,对表达致癌基因RAS的细胞具有选择性。
Ferroptosis是一种独特的铁依赖的非凋亡性细胞死亡形式。Ferroptosis由致命小分子erastin引发,该化合物对癌基因RAS具有选择性。Ferroptosis的激活导致癌细胞的非凋亡性损伤。
在BJ-TERT/LT/ST/RASV12细胞中,erastin引发一种迅速的、氧化的和非凋亡性的细胞死亡过程。Erastin对人癌细胞产生致命性损伤,这些细胞具有参与 RAS-RAF-MEF通路的突变致癌基因HRAS、KRAS和BRAF,erastin是通过调节VDAC(电压依赖性阴离子通道)起作用的[1]。在HT 1080细胞中,erastin诱导的细胞死亡引发了细胞溶质ROS的累积,而且诱导的氧化性死亡是铁依赖性的。Erastin也抑制非依赖性的胱氨酸/谷氨酸逆向转运体XC-系统的活性[2]。

Erastin激活快速、氧化及非凋亡细胞死亡过程。在致癌基因HRAS、KRAS或BRAF突变的人类肿瘤细胞中,Erastin表现出更大的致死性。在携带致癌RAS的细胞中,Erastin导致氧化物质的出现,随后通过氧化及非凋亡机制死亡。在NRAS突变HT-1080纤维肉瘤细胞中,RSL分子Erastin(10 μM)处理以时间依赖性方式增加细胞溶质和脂质ROS的起始。 |
