CP 31398 dihydrochloride是一种强效的p53激活剂,最大耐受剂量为400 ppm [ 2 ]。
肿瘤蛋白p53(p53)是多细胞生物的一个关键蛋白,在预防肿瘤形成中起重要作用。许多研究表明,活化的p53调节p21的表达,p21与G1-S/CDK 复合物(该分子的表达对细胞周期的G1/S过渡期非常重要)结合,并抑制它们的活性[1]。
CP 31398 dihydrochloride是一种强效的p53稳定剂,而且有望与化疗药物联合用药用于癌症的治疗。在9个人肿瘤细胞系中,CP 31398 dihydrochloride导致突变或野生型p53表达的细胞系凋亡(SW480、SKBr3、PA1、U20S、HCT116、Saos-2),并增强化疗药物对细胞的杀伤作用,而对不表达p53的细胞无影响[1]。在4种人头颈部鳞癌细胞系(JHU-O29、UMSCC-22A和Fadu)中,CP 31398 dihydrochloride给药96小时后,可通过累积p53的表达而抑制细胞生长[2]。
在结肠腺癌F344大鼠模型中,联合使用塞来昔布和低剂量的CP 31398 dihydrochloride,可通过增强p53的表达,显著抑制结肠腺癌的发病率(78%)和多样性(90%),以上表明低剂量的CP 31398 dihydrochloride和塞来昔布联合用药有望成为临床治疗结肠癌的方法[3]。
