Tandutinib(MLN518,CT53518)是一种新型的FLT3小分子选择性抑制剂,IC50值为0.22 μM[1].
在体外激酶试验中,Tandutinib(MLN518)抑制FLT3\PDGFR和c-Kit,IC50值分别为0.22 μM\0.20 μM和0.17 μM.此外,Tandutinib(MLN518)可抑制野生型FLT3或W51酪氨酸磷酸化,IC50值为30–100 nM.另外,Tandutinib(MLN518)在FLT3-ITD阳性细胞(Molm-13和Molm-14细胞)中具有抗细胞增殖能力,IC50值为10 nM,但FLT3-ITD阴性细胞(THP-1\KG-1和RS4细胞)对其耐受,需超过1000倍的浓度才可抑制细胞生长[1].
