NSC 66811是一种小分子的鼠双微体2(MDM2)-p53相互作用的抑制剂,Ki值为120 nM[1]。
p53的N端和MDM2的深疏水裂缝可在p53和MDM2之间形成一个短螺旋链。NSC 66811可与这一裂缝的活性位点结合。NSC 66811具有高结合亲和力,Ki值为120 nM,比天然p53多肽强56倍。NSC 66811的立位异构体模仿p53的三个关键残基(Phe19、Trp26和Leu26),介导与MDM2的相互作用。在具有野生型p53的HCT-116人类结肠癌细胞系中,NSC 66811可减少p53、MDM2和p21cip1/waf蛋白的积累。但是,NSC 66811在同基因的HCT-116 p53 /细胞系中对p53、MDM2和p21cip1/waf蛋白水平并无影响[1]。
