神经降压素(NT)可激活G蛋白偶联受体,神经降压素受体1(NTR1)。NT,一种13氨基酸的神经肽,在中枢神经系统与肠包括回肠3和结肠中表达。
神经降压素受体1(NTR1)是一种高亲和力的G蛋白偶联受体(神经降压素,NT)。
体外:由于NT/NTR1信号传导允许miR-133α的表达,NT调节miR-133α表达的机制和研究miR-133α对人NCM460结肠细胞内NTR1运输的作用非常重要。负转录调节蛋白(锌指E-box结合同源异形框1)参与NT诱导的miR-133α上调。miR-133α的结合靶点(MIR-133α沉默或aftiphilin(AFTPH)过表达)降低NTR1向人体结肠细胞质膜的运输而不影响NTR1的内化。AFTPH早期内涵体和反式高尔基体(TGN)集中在未刺激的人结肠上皮细胞。早期内涵体中AFTPH过表达降低NTR1内化。同时,还增加与内涵体和TGN运输途径相关的蛋白的表达。AFTPH过表达和胞内囊泡的脱酸化增加NTR1的表达。这些结果表明,人体结肠上皮细胞中GPCR运输的新机制说明了为什么microRNA,miR-133α通过其下游靶点AFTPH调节NTR1运输。
