免疫球蛋白(Ig)是B细胞的抗原识别分子。Ig分子是由2条相同的重链和2条相同的轻链通过二硫键连接形成的,每条重链与轻链连接,2条重链连接在一起。每个免疫球蛋白重链含有N末端可变区(V)和C-末端恒定区(C),抗原结合位点位于N末端可变区(V),C-末端恒定区是由个别的C区基因编码的,用于确定抗体的亚型,并提供了效应器和发挥信号功能。
IGHM基因编码重链的恒定区,决定了IgM的亚型。Naive B细胞在其表面表达跨膜形式的IgM和IgD。抗体应答期间,通过体细胞重组过程,活化的B细胞可以转换成表达下游重链C区基因。另外,对重链C区序列的可变RNA加工可产生分泌Ig形式,可以作为抗体存在。尽管细胞膜上所有Ig是作为单体存在,但分泌的IgM在血浆中可形成五聚体甚至是六聚体。
