Huwentoxin XVI(HWTX-XVI)由39个氨基酸残基组成,包括6个半胱氨酸。HWTX-XVI是一种源于中华狼蛛O. huwena毒液的新型神经毒素,可特异性阻断N型钙通道。[1]
N型钙通道常在背根神经节细胞体和向脊髓背角突出的突触末端高表达。N型钙通道在控制神经递质释放和将痛觉信号传递到中枢神经系统中扮演重要角色。[2]
电生理学试验表明,在大鼠背根神经节细胞中,HWTX-XVI可特异性抑制N型钙通道。HWTX-XVI可特异性抑制大鼠DRG细胞中GVIA敏感的N型钙通道。[1]
HWTX-XVI可几乎完全阻断大鼠输精管对低频率电刺激的痉挛反应。HWTX-XVI的镇痛效果、无毒性和完全可逆性使之成为开发新的治疗N型钙通道相关疾病药物的工具。[1,2]
