真核起始因子(eIF)是参与真核翻译起始阶段的蛋白质。eIF3结合到核糖体亚基-mRNA复合物,在延伸准备就绪之前,参与阻止大的核糖体亚基与小亚基的结合。在哺乳动物中,eIF3是最大的染色体支架起始因子(scaffolding initiation factor),由13个亚单位组成(a-m),分子量大约为750 kDa,控制40S核糖体亚基在含有5'帽或IRES(内部核糖体进入位点)的mRNA上组装。 eIF3使用来自病毒的eIF4F复合物或IRES定位到靠近40S核糖体亚单位出口处的mRNA链,从而推动转录起始复合物的组装[1]。
位于8q23-q24处的PTK2和EIF3S3基因可能是扩增靶标,和肝癌有关[2]。在晚期前列腺癌中,普遍存在EIF3S3基因的扩增,它可能功能上参与了疾病的进展[3]。最近的研究表明,在前列腺癌中,真核翻译起始因子3亚基3未出现不依赖于c-myc基因的过表达[4]。
