STF-62247可在野生型VHL和VHL缺失的肾细胞癌(RCC)中以HIF不依赖的方式抑制肿瘤生长,IC50值分别为16 μM和0.625 μM.
von Hippel-Lindau(VHL)肿瘤抑制基因的失活可导致大量肾细胞癌(RCC),且与高度血管化和不良预后紧密相关.STF-62247在体外和体内对VHL缺失细胞具有选择性细胞毒性.[1]
体外实验:体外研究证明,STF-62247以HIF不依赖的方式对野生型VHL和VHL缺失的肾细胞癌(RCC)具有选择性细胞毒性和肿瘤生长抑制活性,IC50值分别为16 μM和0.625 μM.此外,STF-62247也可在VHL缺失的细胞中诱导酸化和增加自噬,从而导致细胞凋亡.[1]
体内实验:用于研究STF-62247活性的动物实验遵循机构和国家的指导方针,由斯坦福大学实验动物保护管理专家组批准.基于体内小鼠模型发现,腹腔内注射剂量为8 mg/kg的STF-62247可显著抑制VHL缺失SN12C肿瘤细胞的肿瘤生长.[1]
临床试验:迄今为止尚未进行临床试验.
