F-BL21(DE3)感受态(F-BL21(DE3) Competent Cell)是经特殊方法制备得到的BL21(DE3)感受态,可实现快速转化,大大简化实验流程。BL21(DE3)感受态广泛用于IPTG诱导的蛋白表达实验,其特别适合T7启动子驱动的重组蛋白表达体系,比如最常见的原核表达载体pET系列。同时BL21(DE3)含有大肠杆菌RNA聚合酶,也可用于非T7启动子表达载体(如pGEX,pMAL等)的蛋白表达。
BL21(DE3)感受态的基因型为F- ompT hsdSB(rB- mB- ) gal dcm(DE3)。本菌株缺乏内源性蛋白酶,能有效避免重组蛋白的降解。同时本菌株为被λ噬菌体DE3溶原化得到的BL21衍生菌株,噬菌体DE3中的T7 RNA聚合酶受到lacUV5启动子控制,当外源添加IPTG时可以诱导BL21(DE3)菌株中T7 RNA聚合酶的快速表达,从而驱动重组蛋白的大量表达。本菌株携带ompT突变,该基因缺失可以减少外膜蛋白酶的表达量,提高目的蛋白的表达产量;hsd基因突变可导致菌株内的EcoK或EcoB限制酶活性缺失,从而提高PCR产物的转化效率;dcm基因缺失可以消除甲基化对腺嘌呤或胞嘧啶的影响,从而帮助如携带易受甲基化影响的酶切位点的外源质粒的表达。
本产品提供即用型的BL21(DE3)快速转化感受态,同时提供pUC19质粒作为模板,当使用pUC19质粒(2686bp,AmpR)进行转化时,本感受态转化效率可达1 x 107 CFU/µg以上。本感受态经过特殊方法制备,最快可实现30 s快速转化,大大减少了操作时间,使用十分方便。(注:本产品采用特殊制备方法,肉眼观察为浅棕色,但不影响使用。)
