体外:Zaprinast是一种PDE5和PDE6抑制剂,在适度的高浓度(29 mM)下抑制PDE9 [1]。其表明,通过Zaprinast输注抑制cGMP的水解可增加ANP对排钠的效用,而不引起有害的血压下降。因为已知ANP受体位于肾小球和内髓集合管内,确定肾脏PDE9酶的细胞定位将很有意义[2]。该酶对cGMP显示高特异性,具有cGMP结合位点,其对Zaprinast的敏感性与平滑肌PDE5相似,导致两种酶都被命名为cGMP-PDE。然而,视网膜cGMP-PDE首先被区分为感光体cGMP-PDE,特异性地分布在视网膜,且具有比其他cGMP-PDEs较高的Vmax和Km值,受到G蛋白被调控。 [3]。
