SC-57461A是人重组LTB4的选择性抑制剂,IC50值为49 nM[1]。
LTB4又称白三烯B(4),在炎症中起重要作用,LTB4通过与特定细胞表面受体BLTR1和BLTR2相互作用发挥功能[2]。
SC-57461A是一种强效的LTB4抑制剂,比之前报道的LTA4抑制剂RP64966具有更有效的活性。在PMN细胞中,SC-57461A通过作用于LTB4-BLTR1的相互作用,抑制fMLP和PAF诱导的LT的合成[3]。
在禁食CD大鼠模型中,钙离子载体A23187诱导花生酸生成,口服SC-57461A显著抑制LTB4,ED50值为0.3-1.0 mg/kg。使用AA诱导的耳朵水肿模型中,口服剂量为0.8、4和20 mg/kg的SC-57461A明显抑制水肿[1]。
有研究表明,SC-57461A抑制LTB4A,针对环氧化物水解酶和氨肽酶的Ki值分别为23 nM和27 nM[1]。
