Papain Inhibitor是木瓜蛋白酶抑制剂,是具有序列H-Gly-Gly-Tyr-Arg-OH的肽,MW 为451.4。木瓜蛋白酶是一种存在于木瓜和山木瓜中的半胱氨酸蛋白酶。作用机制是通过His-159裂解去质子化的Cys-25肽键。Asparagine-175有助于His-159的咪唑环定向而发生去质子化。Cys-25攻击肽骨干的羰基碳,使肽的氨基末端游离,形成一个共价酰基酶中间体。这个酶通过水分子脱酰基,并释放肽的羧基端部分。众所周知在免疫学中,木瓜蛋白酶可将Fab(抗原结合的)部分和免疫球蛋白(抗体)的Fc部分裂解[1]。木瓜蛋白酶倾向于在(hydrophobic)-(Arg或Lys)间裂解(不是Val)。Hydrophobic是Ala、Val、Leu、Ile、Phe、Trp或Tyr[2]。木瓜蛋白酶可在细胞培养第一步中分离细胞。也可用作各种enzymatic debriding培养基(特别是Accuzyme)的成分。这些都是一些慢性伤口的护理来清除坏死组织。木瓜蛋白酶也可作为一些牙膏或牙齿增白的成分。
