甘草次酸(enoxolone)是甘草的主要组成部分,同时具有抗病毒和抗真菌活性。它能够羧化 DNA的复制并抑制微生物毒素和酶的生成[1,2]。在大鼠中,18β-甘草次酸有效抑制肝和肾匀浆中11β羟基类固醇脱氢酶 (11β-HSD) 的活性,IC50值分别为0.09 μM和0.36 μM[3]。
在糖皮质激素结合盐皮质激素受体(MR)之前,MR对醛固酮的选择性可通过酶诱导,因此导致竞争性的糖皮质激素失活。11β-HSD-1、11β-HSD-2和11β-HSD-3是11β-HSD的三种酶。11β-HSD-1具有双向活性,Km值在微摩尔范围内,是NADP+依赖性的一种酶。11β-HSD-2仅表现出氧化酶的活性,Km值在纳摩尔范围,是NAD +依赖的一种酶。它与MR在肾脏中共定位[4]。
JEG-3细胞系衍生自人绒毛膜癌。在该细胞系中,11β-HSD-3的酶活性被甘草次酸抑制[4]。
在大鼠中,200 mg/kg的甘草次酸口服给药3小时后,显著抑制了肾脏和肝脏中11β-HSD活性。此外,甘草次酸可以稍微增加循环的皮质酮水平。数据表明,11β-HSD同功酶的抑制活性有一定的差异,11-、24-和30-甘草次酸可能发挥重要作用[3]。
