BVT 2733是一种新型、非甾体和选择性1型11β-羟类固醇脱氢酶(11β-HSD1)的小分子抑制剂。
据报道,11β-HSD1改变胰岛素作用的靶组织中糖皮质激素的作用,并且在葡萄糖体内平衡中起关键作用。
体外:BVT 2733是一种新型1型选择性鼠11β-HSD1抑制剂(Ki为 1 μmol/l),其在200μmol/ l的浓度下不抑制小鼠11β-HSD2型(相当于Ki >33 μmol/l)。因此,基于Ki值,估计BVT 2733对11β-HSD 1型酶的选择性超过对2型酶至少30倍[1]。
体内:与载体处理的小鼠相比, BVT 2733处理小鼠可降低肝脏PEPCK和葡萄糖-6-磷酸酶mRNA、血清胰岛素和血糖浓度。相比之下,BVT 2733的治疗没有改变肝脏11β-羟基类固醇脱氢酶1型mRNA,肝功能标记酶表达包括丙氨酸氨基转移酶、天冬氨酸氨基转移酶和碱性磷酸酶,体重以及日常食物摄入[1]。
临床试验:到目前为止,BVT 2733仍处于临床前研究阶段。
