5-脂氧合酶激活蛋白(FLAP)是一个整合蛋白,在5-脂氧合酶(5-LOX)的活化和白三烯的合成中起重要作用,从而调节免疫应答.
BAY-X1005是白三烯合成的选择性抑制剂.BAY-X1005结合FLAP并抑制5-LOX从细胞质易位到细胞膜[1].BAY-X1005抑制LTB4合成,在人\大鼠和小鼠中分离的PMNL的IC50值分别为0.22\0.026和0.039 μM,抑制小鼠巨噬细胞LTC4的合成,IC50值为0.021 μM [2].
在花生四烯酸诱导的小鼠耳炎症试验中,BAY-X1005抑制髓过氧化物酶活性和水肿的形成,ED50值分别为7.9和48.7[2].
此外,100 mg/kg 的BAY-X1005以剂量依赖的方式将血小板活化因子诱导的小鼠死亡降低了51%.在动物模型中,BAY-X1005抑制LTB4和LTC4的合成,从而降低水肿形成以及血管中炎症和白细胞迁移[3].
