tumor protein p53 binding protein fragment [Homo sapiens]/[Mus musculus]

P53是序列特异性的DNA结合寡聚蛋白，与含p53结合位点的启动子结合，激活转录。p53的活化区域位于氨基末端，目前没有研究报道特异性DNA结合结构域的位置1。

使用酵母双杂交筛选，鉴定到肿瘤蛋白p53结合蛋白（53BP1）可与P53的中心DNA结合结构域相互作用2。类似于乳腺癌易感基因1（BRCA1）3,4， 53BP1增强p53依赖性转录5。有趣的是，53BP1的羧基末端含有串联的BRCA1 COOH基端（BRCT）序列。

在电离辐射条件下，53BP1变得高度磷酸化，可迅速地重新定位于DNA链断裂的位点。在ATM缺陷细胞中，53BP1病灶形成减少，在ATM野生型细胞中，可以通过渥曼青霉素（wortmannin）抑制53BP1病灶形成。此外，使用渥曼青霉素处理的细胞以及ATM缺陷细胞中，不存在辐射诱导的53BP1过度磷酸化。53BP1参与DNA损伤信号传导途径，γ辐射后，可以被ATM调节。
