P53是序列特异性的DNA结合寡聚蛋白,与含p53结合位点的启动子结合,激活转录。p53的活化区域位于氨基末端,目前没有研究报道特异性DNA结合结构域的位置1。
使用酵母双杂交筛选,鉴定到肿瘤蛋白p53结合蛋白(53BP1)可与P53的中心DNA结合结构域相互作用2。类似于乳腺癌易感基因1(BRCA1)3,4, 53BP1增强p53依赖性转录5。有趣的是,53BP1的羧基末端含有串联的BRCA1 COOH基端(BRCT)序列。
在电离辐射条件下,53BP1变得高度磷酸化,可迅速地重新定位于DNA链断裂的位点。在ATM缺陷细胞中,53BP1病灶形成减少,在ATM野生型细胞中,可以通过渥曼青霉素(wortmannin)抑制53BP1病灶形成。此外,使用渥曼青霉素处理的细胞以及ATM缺陷细胞中,不存在辐射诱导的53BP1过度磷酸化。53BP1参与DNA损伤信号传导途径,γ辐射后,可以被ATM调节。
