OAC2是八聚体结合转录因子4的一种激活剂。
已发现八聚体结合转录因子4(Oct4)是胚胎干细胞(ESC)多能性的一种关键调节剂,并且Oct4对重编程过程是至关重要的。
体外:OAC2能够激活Oct4和Nanog报告基因至与OAC1类似的程度,OAC1是对Oct4和Nanog启动子驱动的荧光素酶报告基因表现出最大激活作用的OAC类似物。此外,为了检测4F + OAC1-iPSC和4F + OAC2-iPSC的发育潜力,使用标准胚状体分化方法在体外分化上述细胞。免疫染色结果显示4F + OAC1-iPSC和4F + OAC2-iPSC都能有效分化成特征性平滑肌肌动蛋白(SMA)+中胚层细胞、FoxA2 +内胚层细胞和Tuj1 +外胚层细胞[1]。
体内:为了在动物体内测试4F + OAC2诱导的iPSC的体内多能性,将4F + OAC2-iPSC移植到免疫缺陷的裸鼠中。移植后4至6周,4F + OAC2-iPSC可以有效地产生包含所有三个胚层的衍生物的典型畸胎瘤,例如中胚层的血液、内胚层的肠上皮以及外胚层的表皮[1]。
临床试验:到目前为止,OAC2仍处于临床前研究阶段。
