我们保留了Pro-Leu-Gly-Leu的序列,但在N端标记了高荧光的(7-甲基香豆素-4-) 乙酰基(Mea)基团。Dnp淬灭基团被插入到新的N3-Dnp-t-2,3-二氨基丙酸(Dpa)的P2’位点上[1],因为基质金属蛋白酶偏好芳香族侧链的P2’位点[2,3]。
在非淬灭肽Mca-Pro-Leu中,最大荧光激发波长为328nm,发射波长在393nm附近。在1 M下比较,Mca-Pro-Leu的荧光强度比Mca-Pro-Leu-Gly-Leu-Dpa-Ala-Arg-NH2强130倍。
Mea-Pro-Leu-Gus)-Lrlt-Dpu-AMca-Pro-Leu-Gly-Leu-Dpa-Ala-Arg-NH2的剪切产物是目前所知最敏感的基质金属蛋白酶连续测定的底物,不仅是因其Kcut/Km值比以往报道的物质高,由于Mea基团的荧光比色氨酸强,也能被Dnp基团有效淬灭,其剪切产物能被快速检测。除胶原蛋白酶外,新的底物在与其偏好的大分子底物实验所需的相似酶浓度下,连续检测也能进行。因此,Mca-Pro- Leu-Gly-Leu-Dpa-Ala-Arg-NH2适用于粗制剂中总金属蛋白酶活性的检测。后续对可被特定酶切割的多肽的研发将能满足这些条件下对特定酶的检测[3]。
