钙离子(钙)信号通路在受精中发挥主要作用,精子不仅提供其遗传物质,也使细胞内钙离子浓度升高,这对于完成卵母细胞活化和胚胎发育是必要的。离子霉素是由链霉菌产生的离子载体。工业上,离子霉素通常为游离酸或钙盐。
体外实验:离子霉素是用于研究胞内钙水平的提高和研究跨膜钙转运的工具。它也可以用来刺激胞内产生细胞因子,如干扰素、穿孔素、IL-2和IL-4。这些细胞因子在炎症反应中发挥重要作用[1]。
在体实验:10 μmol/l的离子霉素有效激活卵母细胞的活化和随后的发育效率。二次给予5 μmol/l离子霉素明显改善囊胚发育。在体外或体内小鼠胚胎发育中使用离子霉素无副作用[1]。
临床试验:截至目前,离子霉素仍处于临床前开发阶段。

Applications |
The in vitro growth rate of HT1376 cells was suppressed by ionomycin in a dose- and time-dependent manner. After continuous treatment with 10 μg/ml ionomycin, characteristic apoptotic DNA degradation, as well as marked decreases in the ratios of Bcl-2 to Bax mRNA and protein could be observed in HT1376 cells. |

Applications |
Intratumoral injection of ionomycin into subcutaneous HT1376 tumors reduced the tumorigenicity in nude mice. Furthermore, these in vivo growth-inhibitory effects of ionomycin were significantly enhanced by pretreatment with cisplatin. |
