Fluphenazine是一种多巴胺D1和D2受体抑制剂。
多巴胺D 1和D 2受体免疫组织化学已被用于研究成年大鼠弓形中值隆起复合物的结构,特别是与结节-多发性多巴胺神经元有关。
体外:先前研究表明,Fluphenazine和奋乃静的吩噻嗪诱导细胞活力的浓度依赖性损失, Fluphenazine和奋乃静的EC50分别为1.24和2.76 μM。此外,1.0 μM的Fluphenazine以及1.0 μM和3.0 μM的奋乃静可以抑制黑色素生成并降低小神经炎相关转录因子的含量。此外,Fluphenazine和奋乃静在较高浓度都引起黑色素细胞抗氧化状态的消耗,表明氧化应激诱导[1]。
体内:0.03-0.3 mg/kg剂量的全身性Fluphenazine给药可以有效减弱大鼠神经性疼痛模型中机械性异常性疼痛,其接近于在精神病啮齿动物模型中使用的剂量。对于止痛效应,Fluphenazine能够有效地抑制受损的传入纤维中的异位放电,而不影响来自CCI大鼠的离体DRG神经制备物中动作电位的传播[2]。
临床试验:以商品名Prolixin出售的Fluphenazine是临床上用作治疗慢性精神病如精神分裂症的抗精神病药物。此外,Fluphenazine在效力上约等同于低效抗精神病药。
