FLI-06是一种新型的Notch信号通路抑制剂,EC50值为2.3 uM[1].
Notch信号通路在许多细胞的命运决定中起重要作用,其异常活性导致癌症和发育障碍[1].
用FLI-06处理HeLa NotchΔE-eGFP细胞,导致NotchΔE-eGFP累积和NICD-eGFP生成减少.将FLI-06洗脱后,表型在1–4 h内完全可逆,这表明FLI-06对细胞不是极性毒性的[1].在 FLI-06 处理的细胞中,Aβ 分泌显著降低而不是APPCTF 积累,这表明FLI-06 在α-分泌酶和β-分泌酶裂解的上游起作用.对HeLa细胞进行免疫荧光分析显示, FLI-06 引起高尔基体的完全中断,这可通过干扰早期分泌途径的膜转运或通过分解微管17引起.FLI-06抑制ER消退,也引起tubule-to-sheet表型,且彼此相关[1].
