Diphenyleneiodonium chloride (DPI)是GPR3的激动剂,EC50值为1 μM[1]。另外,DPI可以抑制NADH氧化酶(NOX)、一氧化氮合成酶和细胞色素P450还原酶[2][3][4]。
G蛋白偶联受体-3(GPR3)是G蛋白偶联受体(GPCR),具有基本的G(s)信号传导活性,激活cAMP,主要表达于哺乳动物的脑和卵母细胞中[1]。
DPI是GPR3的激动剂。在稳定表达GPR3的HEK293细胞中,DPI增加细胞内cAMP的水平,且不依赖于NOX的抑制作用。 DPI激活表达GPR3的细胞的腺苷酸环化酶。在共表达β-arrestin2和GPR3的HeLa细胞中,DPI增加钙的流动、GPR3受体脱敏和β-arrestin2的膜定位[1]。DPI抑制HeLa细胞中NADPH氧化,EC50值为0.1 μM[2]。在小鼠巨噬细胞中,DPI不可逆的完全抑制一氧化氮合酶,而NADP+、NADPH或2'5'-ADP可阻断这种抑制作用。此外,DPI抑制乙酰胆碱诱发的去甲肾上腺素预收缩兔主动脉环的松弛,IC50值为300 nM[3]。DPI不可逆地抑制NADPH细胞色素P450氧化还原酶,Ki值为2.8 μM[4]。
在大鼠中,DPI(10-5 mol/kg)抑制乙酰胆碱引起的降压反应[5]。
