Cinnarizine是一种钙通道阻断剂。
钙通道阻滞剂是破坏钙通过钙通道移动的药物,通常用作降压治疗以降低高血压患者的血压。
体外:先前研究发现,cinnarizine可以抑制动脉肌球蛋白P-轻链的磷酸化和动脉肌动球蛋白超沉淀。此外,研究发现perhexiline和Cinnarizine对动脉超沉淀和磷酸化的伴随抑制与W-7相似。这种抑制作用的特征在于pCa超沉淀右移,最大活性降低以及外源钙调素的减弱[1]。
体内:先前动物研究表明,当cinnarizine与丙戊酸钠联合用药时,在MES癫痫发作模型中获得增强效应。然而,在PTZ诱导的癫痫发作中,当硝苯地平与丙戊酸钠联合用药时获得增强效应[2]。
临床试验:对具有良好表征的外周或中枢前庭紊乱的四十位患者进行双盲对照研究,以评价cinnarizine的抗眩晕效应。电子震动结果显示,用cinnarizine和安慰剂治疗的患者之间没有明显的可检测差异,当cinnarizine剂量增加至150 mg时,在5个健康志愿者中仅观察到对前庭眼球震颤的临时抑制作用。此外,研究发现cinnarizine通常具有良好的耐受性[3]。
