Chetomin,一种螺卷毛壳霉的抗生素代谢物,是HIF-1的抑制剂,能削弱HIF-1转录。最近,它作为癌症化疗剂备受关注[1]。
HIF-1(缺氧诱导因子1)是响应于细胞环境氧变化的转录因子[2]。HIF-1复合物属于转录因子碱性螺旋-环-螺旋家族中的PAS亚家族,由α和β亚基形成异源二聚体[3]。HIF-1通过与启动子的HIF效应元件结合,可上调某些基因,如:糖酵解酶,血管内皮生长因子,以促进细胞在缺氧条件下的存活率。研究表明,在正常情况下,NF-κB是HIF-1α的直接调控元件,但在低氧条件下HIF-1α仍保持稳定,机制未明[4]。
Chetomin通过干扰HIF-1与其转录辅激活因子p300的相互作用,从而选择性地抑制HIF-1的活性。在稳定转染携带缺氧反应元件5HRE-hCMVmp启动子质粒的早期传代人纤维肉瘤HT1080细胞中,Chetomin预处理后,对EGFP荧光信号的抑制呈现出剂量和孵育时间依赖性。用150 nM Chetomin预处理4小时呈现出最大抑制效果,表明Chetomin对HIF依赖性转录的抑制作用。此外,150 nM Chetomin降低处于缺氧条件下的HT1010细胞中VEGF和CA9的表达。相对于常氧的条件,Chetomin增加缺氧条件下细胞的存活率,表明Chetomin可以增强在严重缺氧条件下的放射治疗疗效[5]。
