AS 1269574,(2-[2-(4-溴苯基)-6-甲基嘧啶-4-基]氨基乙醇),可在正常小鼠中诱导葡萄糖刺激的胰岛素分泌(GSIS)及提高葡萄糖耐受。AS 1269574是GPR119的激动剂,EC50值为2.5 μM[1][2]。
GPR119在胰腺β细胞中高表达。该受体通过提高胞内cAMP浓度,增强GSIS效果[2]。
以1或10 μM的AS 1269574处理20 min,16.8 mM葡萄糖预处理的小鼠胰腺MIN-6 β细胞的胰岛素分泌与DMSO溶剂组相比显著提高。相比之下,AS 1269574不可影响低浓度葡萄糖(2.8 mM)预处理的MIN-6细胞的胰岛素释放[2]。
在正常小鼠中,以100 mg/kg的AS 1269574单一处理可显著降低2 h后(AUC0-2h)的血糖的曲线下面积(AUC)。AS 1269574相比溶剂对照可显著提高小鼠的血浆胰岛素AUC0-2h。单一口服100 mg/kg的AS 1269574不能显著降低禁食或喂食小鼠的葡萄糖AUC,表明AS 1269574不具有降血糖作用[2]。
