ZLN005是一种新型的PGC-1α转录调控子[1].
过氧化物酶体增殖体激活受体γ共激活因子-1α(PGC-1α)是一种诱导型共调节因子,可调节能量代谢.PGC-1α调控葡萄糖和脂肪酸代谢\线粒体生物发生等等.PGC-1α失调与胰岛素抵抗和2型糖尿病有关[1].
ZLN005是一种新型的PGC-1α转录调控子.在PGC-1α启动子报告试验中,ZLN005可有效地抑制荧光素酶.在L6肌管中,ZLN005可剂量依赖地增加PGC-1α的mRNA水平.ZLN005(10 μM)可提高细胞色素c氧化酶5b(cox5b)\乙酰辅酶A氧化酶\雌激素相关受体α(ERRα)\NRF1和GLUT4的mRNA水平.ZLN005(20 μM)可以剂量依赖的方式分别提高1.8倍和1.28倍的葡萄糖吸收和棕榈酸氧化.在转染PGC-1α siRNA的L6肌管中,ZLN005刺激的棕榈酸氧化和PGC-1α基因表达被阻断.在L6肌管中,ZLN005可提高AMPK和乙酰辅酶A羧化酶(ACC)的磷酸化[1].
在db/db小鼠中,ZLN005可减少RER,表明转换为脂肪酸利用.ZLN005可显著降低空腹血糖和随机血糖水平.另外,ZLN005可提高葡萄糖清除率和丙酮酸耐受,降低胰岛素抵抗[1].
