YYA-021是小分子CD4类似物,能抑制HIV侵入[1].
CD4是一种细胞表面蛋白.1型人免疫缺陷病毒(HIV-1)通过病毒包膜糖蛋白gp120与细胞表面CD4的相互作用,进而侵入靶细胞.CD4类似物抑制gp120与CD4的相互作用,由此抑制HIV侵入[1].
YYA-021是小分子CD4类似物,能抑制HIV侵入.YYA-021完全抑制gp120-CD4相互作用[1].在感染猿猴人类免疫缺陷病毒分离株MNA(SHIV MNA)或HIV-1 MNA假病毒的TZM-bl细胞中,YYA-021抑制SHIV MNA和HIV-1包膜糖蛋白侵入.KD-247 (anti-V3 loop mAb,50 μg)中和50% 的HIV-1 MNA.然而,20 μM YYA-021存在的情况下,KD-247 (<0.05 μg)中和50%的HIV-1 MNA,表明YYA-021和KD-247具有协同中和作用.YYA-021存在的情况下,SHIV MNA对IgG更加敏感[2].
在Jcl:SD大鼠中,YYA-021.HCl的最大耐受剂量为2.5 mg.尾静脉注射的YYA-021基于其疏水性,表现出广泛的组织分布.在恒河猴中,YYA-021.HCl的最大耐受剂量为35.3 mg [1].
