TCS3035是GPR35的激动剂,对大鼠和人GPR35同源物显示出显著较高的效力,pEC50值分别为5.13和5.86[1]。G蛋白偶联受体(GPCRs)是最大的和最广泛研究的跨膜多肽。GPR35是特征较差的G蛋白偶联受体,在免疫调制、胃功能和胰岛素分泌的调节中起重要作用。GPR35主要在胃肠道和免疫组织中表达。色氨酸代谢物犬尿酸被认为是GPR35的内源性配体[1]。
体外实验:在FLAG-hGPR35-eYFP 或 FLAG-rGPR35-eYFP 中,3.36位点的保守精氨酸突变为丙氨酸导致TCS3035的激动剂功能完全丧失[1]。此外,TCS3035诱导的GPR35内化与受体-β-arrestin-2相互作用的BRET实验中TCS3035的效力相关。跨膜结构域III的3.32位点酪氨酸突变为丙氨酸消除了响应于TCS3035的β-arrestin-2的募集[1]。
