神经激肽3(NK-3)受体是G蛋白偶联受体超家族的成员,作为速激肽的受体而起作用。激活的NK-3受体通过激活磷脂酶C,引起磷酸肌醇4,5二磷酸(PIP2)分解成1,4,5三磷酸肌醇(IP3)和二酰基甘油[1]。
在表达hNK-3受体的中国仓鼠卵巢(CHO)细胞中,SB 222200抑制神经激肽B(NKB)与细胞膜的结合,Ki值为4.4 nM。在表达hNK-3受体的HEK 293细胞中,SB 222200拮抗NKB诱导的Ca2+动员,IC50值为18.4 nM[1]。
在小鼠模型中,SB-222200剂量依赖性地抑制senktide(NK-3受体的选择性激动剂)诱导的行为反应(快速摇头和尾鞭),ED50值为5 mg/kg,该抑制效应与SB-222200在脑内的浓度显著相关[1]。用SB 222200处理成年雄性CD-1小鼠,当用可卡因挑战时,小鼠具有显著增强的多动行为,表明NK-3受体的阻断增强多巴胺介导的多动行为[2]。
