Rotenone(鱼藤酮)是线粒体复合物I电子传递链的抑制剂,IC50值为1.7-2.2 μM。
电子传递链(ETC)由一系列转移电子的化合物组成,可以将电子从电子供体传递到电子受体,并跨膜转运质子(H+)。质子梯度驱动ATP的合成,复合物I是产生超氧化物的主要位点之一。
鱼藤酮是线粒体复合物I电子传递链的抑制剂。在转化细胞系HEK293和癌细胞系U87中,鱼藤酮(50 μM)以剂量依赖的方式诱导活性氧(ROS)介导的细胞死亡,死亡率分别为30%和40%。此外,鱼藤酮显著诱导细胞自噬的形成[1]。在SH-SY5Y细胞中,鱼藤酮诱导caspase依赖的细胞凋亡。鱼藤酮也诱导p38 MAP激酶、c-Jun和JNK的磷酸化,表明p38和JNK途径的活化[2]。在分化的SH-SY5Y神经母细胞瘤细胞中,鱼藤酮(50 nM)诱导60%的细胞死亡,减缓线粒体的运动,但鱼藤酮不诱导类似Lewy小体的形成[3]。
