Naphthoquine phosphate

对各种恶性疟原虫株的IC50值为0.1 ~ 5.2 nM[1]。

疟疾仍然是具全球毁灭性影响的疾病，每年导致80多万人死亡，而且绝大多数是5岁以下儿童。Naphthoquine phosphate是中国发现的抗疟疾药物。

体外：Naphthoquine phosphate对各种恶性疟原虫株具有良好的抗疟疾活性。此外，以往研究表明，Naphthoquine对氯喹耐药株也有效[1]。

体内：Naphthoquine phosphate与青蒿素联合用药对抗恒河猴诺氏疟原虫的研究结果表明，Naphthoquine phosphate与青蒿素联合用药优于单一用药，组合的最佳比例为1：2.5[2]。

临床试验：口服给予无病发症恶性疟疾成年患者单剂量的药物组合（400 mg Naphthoquine + 1000mg 青蒿素）。平均退热时间和原虫转阴时间分别为18.2 ± 8.6 h和34.6 ± 14.3 h。此药物组合具有有效的抗寄生虫作用，临床有效率为98.1%（54例中有52例）。此药耐受性良好，在患者中没有检测到任何不良反应[3]。
