KNK437是亚苄基内酰胺化合物,抑制应激诱导的HSP(热休克蛋白)合成.
热休克蛋白(HSPs)是分子伴侣,在不同的细胞应激下其表达增加,如温度升高.它在调节蛋白质折叠中发挥至关重要的作用,与癌症和心血管疾病相关.
在人结肠癌细胞(COLO 320DM)中,KNK437抑制耐热的获得,以剂量依赖性方式诱导各种热休克蛋白.KNK437也会阻断亚砷酸钠诱发的耐热性获得[1].
KNK437在体内毒性较低.在移植SCC VII 细胞的C3H/He小鼠中,肿瘤中KNK437的浓度逐渐增加,腹膜内注射6小时后达到峰值.44℃高温处理10分钟,8小时后Hsp72被合成.在热刺激之前6小时KNK437给药可抑制其合成[2].
