颗粒酶B是一种细胞凋亡诱导因子,表达于自然杀伤(NK)细胞以及细胞毒性淋巴细胞(CTLs)颗粒和细胞毒性T细胞中,在诱导炎症和细胞外基质重塑中起着重要作用[1].
Z-AAD-CH2Cl(Z-AAD-CMK)是一种细胞渗透性的和不可逆的颗粒酶B抑制剂.在表达NK细胞表面抗原的人C-28/12软骨细胞中,Z-AAD-CMK呈剂量依赖性地抑制软骨细胞对K562细胞的细胞毒作用,该作用依赖于颗粒酶B[1].在口腔鳞状细胞癌细胞系OSC-3中,与对照组相比,Z-AAD-CMK减少白介素-2激活淋巴细胞(LAK细胞)增强的caspase-3活性(约70%),并抑制约20%的 DNA片段.与对照组相比,Z-AAD-CMK也抑制LAK细胞诱导的活性氧中间体(ROI)增加(约50%)[2].在Tc1细胞中,Z-AAD-CMK抑制细胞凋亡[3].
