蛋白酶活化受体2(PAR2)亦称凝血因子II(凝血酶)受体样1(F2RL1)或G蛋白偶联受体11(GPR11),在血管组织和高血管器官中表达,参与血管张力调节.PAR2在血管炎症和伤口愈合过程中起重要作用[2].
AC 264613是选择性PAR2激动剂,对其它PARs没有效应.对HEK 293T细胞(野生型PAR2)和KNRK细胞(转染人PAR2受体)进行刺激PI水解和Ca2+动员实验,AC 264613表现出一定的效应,pEC值分别为6.9和7.0 [1].利用QuickChange诱变的方式,在含有ECL2 突变Q233E\E232R\E232R/Q233R\F240S和F240S/S37P受体的NIH 3T3细胞中,AC 264613处理的PAR2 F240S受体在多种功能端点组成型表达,而对E232和Q233突变体没有作用[2].
在SLIGRL-NH2或胰蛋白酶诱导的急性热伤害和足趾肿胀的雄性SD大鼠模型中,趾内给予低至30 ng AC 264613能够诱导后趾水肿和热痛觉过敏,并表现出剂量依赖性促疼痛效应[1].
