A-967079是瞬时受体电位A1(TRPA1)受体的拮抗剂,对大鼠和人TRPA1受体的IC50值分别为289 nM 和67 nM [1]。
瞬时受体电位(TRP)离子通道是一个较大的家族。包含有TRPV、TRPM、TRPC、TRPN、TRPP和TRPML。TRPA1是这个家族的一部分,在小型和中型的肽能初级感觉传入神经元中也表达TRPV1 [1]。
在WT TG神经元中,A-967079减弱Ca2+信号,在Trpv4-/- TG神经元中,A-967079导致响应于福尔马林的Ca2+信号进一步减少。A-967079可以快速的阻断TRPA1,将应答细胞的百分比从27.7%降低到15.8%,在Trpv4-/- TG神经元中,与TRPA1抑制剂共同作用的效果仅为3.2%[2]。
WDR神经元的自发活动表明中枢敏化,可能引发非诱发或持续性疼痛。在未受伤和CFA炎症大鼠中,30 μmol/kg 剂量的A-967079静脉内注射,导致同侧后爪的有害刺激引起的宽动态范围(WDR)和感受伤害特异性(NS)神经元的响应降低。同样地,在OA和OA-假大鼠中,A-967079减少膝关节高强度机械刺激引发的WDR神经元响应。在CFA炎症大鼠中,A-967079减少低强度机械刺激(10 g von Frey hair)诱导的WDR神经元应答,但在未受伤的大鼠中没有作用。此外,在CFA炎症大鼠中,A-967079注射降低WDR神经元的自发活动,而在未受伤、OA、OA-假动物中没有此种现象[1]。
