通过筛选天然产物来源以发现角鲨烯合酶的抑制剂,两个相互独立的研究组发现了zaragozic acids (ZAs),一种含有新型4,6,7-三羟基-2,8-二氧代二环[3.2.1]辛烷-3,4,5-三羧酸核的真菌代谢物家族。
体外:Zaragozic acid A及其两种类似物Zaragozic acid B和Zaragozic acid C分别从未鉴定的无菌真菌培养物、Spororniella intermedia和Leptodontium elatius发现。据报道,Zaragozic acid A、B和C是三种强效的大鼠肝角鲨烯合酶竞争性抑制剂,Ki值分别为78pM、29 pM和45 pM。此外,Zaragozic acid A、B和C也可以抑制Hep G2细胞中的胆固醇合成[1]。
体内:在小鼠模型中发现,Zaragozic acid A是急性肝胆固醇合成的一种抑制剂(50%抑制剂量为200 μg/ kg)。角鲨烯合酶的体内抑制伴随着标记物从[3H]甲羟戊酸向二磷酸法呢酯、法呢醇以及有机酸的积累。总之,这些数据表明,zaragozic acids是一类具有高胆固醇血症治疗潜力的新型治疗剂[1]。
临床试验:到目前为止,zaragozic acid A仍处于临床前研究阶段。
