6 - 羟基多巴胺(CAS 28094-15-7)是一种儿茶酚胺能神经毒素,被广泛用于在帕金森病啮齿类动物模型中诱导多巴胺能损伤及帕金森病症状。
向黑质、内侧前脑束(medial forebrain bundle, MFB)或纹状体分别注射剂量为 9 μg、6 μg 和 18 μg 的 6-OHDA,会导致小鼠出现运动功能障碍,并使其脑内酪氨酸羟化酶免疫反应阳性神经元显著减少。研究表明,对大鼠进行脑池内给药(200 μg 6-OHDA)可降低其脑内多巴胺与去甲肾上腺素水平。此外,浓度为 50 μM 的 6-OHDA 可诱导原代大鼠皮质细胞及大鼠肾上腺 PC12 细胞发生凋亡。当 6-OHDA 浓度为 75 μM 时,可呈时间依赖性激活半胱天冬酶 - 3、-8 和 - 9(caspase-3、-8、-9),升高 PC12 细胞内活性氧(reactive oxygen species, ROS)水平;且将该浓度 6-OHDA 立体定向注射至大鼠右侧内侧前脑束(MFB)时,可诱导其黑质发生凋亡。
