JMS-17-2 是一种有效的选择性CX3CR1 小分子拮抗剂 (IC50: 0.32 nM)。 它能削弱乳腺癌细胞的转移和定植。 CX3CR1 是 CX3C 趋化因子 fractalkine (CX3CL1) 的受体。 研究表明,趋化因子受体 CX3CR1 能驱动癌细胞进入骨骼,激活正常细胞和癌细胞中的促生存信号通路,促进细胞存活。JMS-17-2对暴露于50nM Fractalkine的SKBR3乳腺癌细胞的ERK磷酸化,能产生剂量依赖性抑制。 SCID 小鼠移植了 MDA-231 细胞,并在 IC 注射后的第一周随机分配并给予载体或 10mg/Kg 的JMS-17-2 腹腔注射,每天两次,持续三周。 结果表明,它既能防止循环肿瘤细胞转化为播散性肿瘤细胞,又能抑制骨骼和内脏器官现有肿瘤数量和大小的扩大。
