Hec1是一种原癌基因,参与纺锤体检验点信号,在多种人类癌症中过表达.Nek2是一种丝氨酸/苏氨酸蛋白激酶,可磷酸化Hec1,这对其有丝分裂功能和细胞存活至关重要[1].
INH6是一种有效的Hec1/Nek2抑制剂.INH6可通过导致染色体错误分离和细胞死亡的蛋白降解,抑制Hec1/Nek2功能.在MDA-MB468和MDA-MB231人类乳腺癌细胞系\HeLa人类宫颈癌细胞系和K562人成红细胞性白血病细胞系中,INH6具有显著的抗增殖活性,IC50值分别为2.1\1.7\2.4和2.5 M.在HeLa细胞抽提物中,INH6耦联基质可选择性地与细胞Hec1共沉淀,表明INH6结合细胞Hec1.在Hela细胞中,INH6(6.25 M)可在8-11 h减少50%的Nek2.此外,INH6随时间进展可略微降低Hec1.另外,INH6可增加具有多极纺锤体配置的有丝分裂细胞数.在表达染色体标记蛋白H2B-GFP的HeLa细胞中,INH6可增加染色体错配.在HeLa细胞中,INH6可诱发垂死细胞发生积极的形态学变化,并诱导20%的细胞凋亡[1].
