产品描述
戊酸倍他米松(Betamethasone Valerate,BV或BET),也称为他米松17-戊酸酯[2],是一种常用的抗炎皮质类固醇,用于制造可用于局部的皮肤病学药物[1,3]。在正常大鼠中,它的IC50 为3.4 ± 0.5 nM[4]。BV可降低体外葡萄糖-6-磷酸脱氢酶(G-6-PDH)活性[5]。
G-6-PDH(EC.1.1.1.49)是催化戊糖磷酸途径第一步骤的酶[6]。
在人皮肤组织匀浆中,使用含有5%二甲基甲酰胺的溶液37℃温育120分钟,随后使用4.6 mM/l的BV,可以减弱G-6-PDH的活性,将G-6-PDH的浓度从约55 mU/ml减少到9 mU/ml [5]。
与每天80 mg剂量的色甘酸钠(SCG)相比,在需要长期使用支气管扩张剂的常年哮喘的儿童中,使用倍他米松戊酸酯(BV)800 μg每天治疗,连续使用4周后,儿童所需的支气管扩张药显著较少,并伴随较少的症状和较高的日常呼气流速[7]。在稳定、轻度、中度牛皮癣病人中,与煤焦油霜(10%LCD)相比, 2周清洗期之后,使用0.1%的戊酸倍他米松乳膏治疗,每天两次,连续6周,PASI的得分与基线相比明显减少。戊酸倍他米松的最终效果得到了显著的清除和改进[8]。
