NCH 51是组蛋白去乙酰化酶的新型有效抑制剂[1]。
去乙酰化酶(HADC)可以除去组蛋白中ε-N-乙酰赖氨酸上的乙酰基,使组蛋白更紧密地包裹住DNA,阻止转录。
在NCI-H460和MDA-MB-231细胞中,NCH-51抑制细胞生长,EC50值分别为2.1和4.4 μM,此外,它可以抑制各种癌细胞系的增殖,EC50值范围为1.1-9.5 μM[1,2]。在HCT 116细胞中,NCH-51以剂量依赖性方式增加乙酰化组蛋白H4和p21WAF1/CIP1的水平[2]。在一系列淋巴癌细胞系中,NCH-51诱导细胞凋亡。在U266细胞中,NCH-51活化caspase-3、-8和-9。另外,NCH-51增加p21表达,降低抗凋亡分子的表达,如survivin、bcl-w 和c-FLIP。在蛋白水平上,NCH-51增加过氧化物酶1 和2、谷胱甘肽-S-转移酶(glutathione S-transferase)和活性氧(ROS)的水平[3]。在HIV-1潜伏感染的细胞中,NCH 51增加组蛋白超乙酰化,减少HDAC1占用,在HIV-1启动子上募集正向转录因子,增加HIV-1的表达[4]。
