CEP-1347,也称为KT 7515,是c-Jun N-端激酶(JNK)信号通路的抑制剂,在大鼠胚胎运动神经元中对JNK1活化的IC50值为20 ± 2 nM [1]。
JNK途径也被称为应激活化蛋白激酶(SAPK)途径,是信号级联之一,介导各种应激刺激的凋亡性死亡。JNK的磷酸化激活对体内损伤和体外营养因子撤出后神经元细胞死亡发挥重要作用[2]。
CEP-1347诱发神经元的存活。在未经治疗的细胞培养物中,平板接种后JNK1的活性在24小时内增加了约四倍。500 nM的CEP-1347处理15分钟后, JNK1的活性急剧下降到对照水平的约50%。在接下来的24小时,JNK1的活性继续下降。在富含培养物的运动神经元中,不断增加CEP-134的浓度,在22小时对JNK1活性的IC50为21 ± 2 nM,在第5天对细胞存活的EC50为20 ± 2 nM [1]。
CEP-1347可影响噪音引起的听力损失。有数据表明,对照组和噪音组之间,噪声干扰前2天的听觉阈值没有显著差异。噪声干扰2天后,所有豚鼠的听力阈值发生改变。噪音处理后第6天,与对照组相比,CEP-1347组的阈值改变显著更低。噪音处理2周后,两组之间的差异变得更加明显[2]。
