Vialinin A是泛素特异性肽酶(USP)的一个有效的抑制剂,对USP4、USP5/异肽酶T(IsoT)和泛素C末端水解酶L1去泛素化酶(UCH-L1 DUB)的IC50值分别为1.5 μM、5.9 μM和22.3 μM[1]。
USPs是去泛素化蛋白的一系列蛋白酶。泛素蛋白的修饰作用可以改变它在细胞中的功能、定位和命运。泛素水解酶在免疫和感染中起着重要的作用[2]。
在RBL-2H3细胞中,Vialinin A抑制肿瘤坏死因子(TNF-ɑ)的产生和释放,并抑制USP5的酶活性。在USP5 siRNA-敲除RBL-2H3细胞中,Vialinin A以剂量依赖的方式抑制TNF-ɑ的产生和释放,TNF-ɑ的mRNA表达量也显著降低[2]。在RBL-2H3细胞中,Vialinin A抑制TNF-ɑ的产生的IC50值为0.09 nM。Vialinin A也可抑制白介素4、已糖氨酶和单核细胞趋化蛋白1的释放,表明它具有抗过敏活性[3]。
