MPC-6827(Azixa)是一种小分子微管去稳定剂, 与β微管蛋白的秋水仙碱结合位点(或附近位点)相结合[1]。
微管蛋白是一种异源二聚体,由一个α单体和一个β单体组成,可通过近紫外辐射被[3H]秋水仙碱共价标记。大多数标记出现在β微管蛋白中。秋水仙素以一个化学计量与微管蛋白结合,亚化学计量地抑制微管组装。秋水仙碱与微管蛋白的结合具有伪不可逆动力学,一个快速的步骤紧跟一个缓慢的步骤,参与配体与微管蛋白构象的变化。反过来,微管蛋白促使秋水仙碱的托酚酮基团的荧光特性[2]。
MPC-6827是一种小分子微管去稳定剂,引起有丝分裂停滞和细胞死亡。MPC-6827干扰微管动力学,导致分裂细胞在细胞周期G2-M期的停滞,最终导致凋亡性细胞死亡[1]。
在体内,MPC-6827显著抑制多种经皮下植入肿瘤细胞系的生长。在人卵巢癌异种移植模型中,MPC-6827也是一种血管破坏剂(VDA)。在小鼠异种移植模型中,MPC-6827与carboplatin具有协同效应。而且,在颅内植入人胶质母细胞瘤细胞系(D-54)的无胸腺裸鼠中,MPC-6827抑制肿瘤生长[1]。
