ML239是最强的乳腺癌干细胞抑制剂,其IC50值为1.16 μM[1]。
ML239选择性地抑制HMLE_sh_ECad细胞,其IC50值为1.18 μM。与对照组相比,ML239对HMLE_sh_ECad细胞具有23倍以上的选择性。此外,ML239对另外一种癌干细胞样细胞系HMLE_shTwist以及乳腺癌细胞系MDA-MB-231显示出毒性。在HMLE_sh_GFP细胞中,给予ML239能在不同程度上调节5个基因(ATP6V0C、PKM2、PPDPF、RPL23和SERINC2)。使用经ML239处理后的细胞进行基因表达研究,结果表明,相对于等基因对照系,HMLE_sh_ECad细胞系中的NF-κB途径在基因表达水平上发生了改变。ML239对另外一种癌干细胞样细胞系(HMLE_Twist)和乳腺癌细胞系(MDA-MB-231)也显示出选择性毒性。类似于在HMLE_sh_ECad细胞系中观察到的结果,ML239具有强效毒性,能抑制HMLE_Twist,其IC50值约为0.1 μM。同时,ML239对乳腺癌细胞系MDA-MB-231也显示出有效的毒性(IC50 = 2.81 μM)[1]。此外,ML239使NF-κB、MAPK以及炎性细胞因子途径在基因表达水平上发生改变。
