LJI308是RSK的强效选择性抑制剂。P90的核糖体S6激酶(RSK)组成了一个丝氨酸/苏氨酸激酶,其在各种人类癌症中均有所表达。RSK是ERK胞外信号调节激酶的胞质底物,参与细胞存活、增殖和细胞极性的直接调节。以前的研究已经证明,RSK通路对于癌症干细胞的生长和增殖至关重要[1,2]。
LJI308对RSK1、2和3的IC50值分别为约0.004 - 0.013 mmol/L。10 μM 的LJI308几乎可以100%的结合RSK2。LJI308与RSK1、RSK3、RSK4的N-末端结合程度类似。LJI308抑制S6K1 的IC50为0.8 mM。在MDA-MB-231和H358细胞系中,1和10 μM的LJI308对于集落形成和细胞生长的抑制效果具有不同灵敏度[1]。在HTRY-LT细胞系中,LJI308(1-10 μM)给药4天或8天后,会降低高达90%的细胞活力,对非致瘤性HTRZ细胞几乎没有影响[2]。
LJI308是RSK抑制剂,在三阴性乳腺癌(TNBC)患者中,可以与常规化疗组合治疗,从而克服耐药性并提高存活率[2]。
