ID-8是一种DYRK小分子抑制剂.在HES2细胞中,ID-8(0.5 μM)可提高1.1%的hESC存活率.ID-8和Wnt3a联合使用可提高1.7%存活率,并能完全抑制Wnt诱导的形态学分化.并且,ID-8可显著降低Wnt诱导的分化标记基因GATA6\GSC\SOX17和CDX2的表达.在hESCs中,ID-8可通过抑制DYRK,提高Wnt介导的hESC增殖和存活.并且,DYRK家族是ID-8的直接靶标[1].在胚胎干细胞(ESC)中,ID-8(10 μM)可在无MEF\血清或LIF的培养物中刺激增殖.ID-8诱导的ESC具有形成所有三种胚层衍生组织的能力,如肝脏\神经元和肌肉.并且,ID-8可提高ESC分化以及Nanog\Sox2和Rex-1水平[2].在无血清培养基中培养的小鼠ESC中,ID-8保留了其自我更新能力和多能性[3].

应用 |
在无血清培养基中培养的D3ES胚胎干细胞(ESC)中,ID-8能够使ESC在未分化状态下增殖至少30天。维持在补充有ID-8的培养基中的细胞可以分化成各种细胞类型,例如神经元,肌肉和肝脏。从长期培养物中去除ID-8导致ESC分化。 ID-8通过Sox2-Oct3/4激活,维持Nanog基因表达,可逆维持ESCs的长期培养。 |
